Abstrakt:
Biogenic amines (BA) are important and irreplaceable biological molecules for the human organism, however, their physiological level for organism is limited. If there is a higher level of biogenic amines in the body they can be toxic for the organism. Currently, various different methods are involved in BA determination including high-performance liquid chromatography (HPLC), thin-layer chromatography (TLC), capillary electrophoresis (CE), ion-exchange chromatography (IEC), flow-injection analysis (FIA) and ion mobility spectrometry (IMS). Generally, the electrophoresis and chromatography are the most used techniques for determination of BA. Both techniques have the ability to load a large volume of samples and to concentrate the analyte in the sorbent. However, the disadvantage is that after the elution of analyte together with solvent only a small amount is available for the detection. To improve the selectivity and sensitivity of detection, some laborious and time-consuming pre-treatment steps are required. Replacement of current laboratory analytical procedures for BA determination by some easier and maybe faster detection is the current challenge. For this purpose, the utilization of specific biosensors can be beneficial. This diploma thesis will be a new approach for the detection of biogenic amines by using electrochemical biosensors modified with selective peptides for specific BA is presented. The aim of this thesis is to design and prepare a sensitive and selective biosensor for the detection of three main biogenic amines (histamine, cadaverine and putrescine). The novelty of this project lies in the utilization of synthetic peptides as highly specific bio-recognition elements and gold nanoparticles as a base for enhancing the sensitivity.
