Abstrakt:
Plants need to adapt to an ever-changing environment, and one of the main stimuli driving this adaptation is the intensity of the photosynthetically active radiation. During the early stages of seedling development, the main form in which small herbaceous plants like Arabidopsis thaliana can affect their accessibility to light supply is by modulating their hypocotyl elongation. This elongation is achieved almost entirely by cell elongation, without any cell division being involved, and is controlled by phytohormones. In this study, we will try to deepen our knowledge about how phytohormones control the cell expansion during hypocotyl elongation in Arabidopsis thaliana seedlings. For that purpose, the main approach will be the use of different microscopy techniques to visualise and quantify cell expansion in different areas of the hypocotyl, and at different times of plant development. We will supply different concentrations of various phytohormones (namely, cytokinins and auxins) and will study their effect in the aforementioned parameters. In addition, we will employ mutants to visualise the cell cytoskeleton and study the effect of each phytohormonal treatment on cytoskeletal organization; a trait that is closely related to cell expansion. Objectives: 1. Learn how to handle in vitro cultures and microscopy techniques 2. Study the effect of different auxins (natural and synthetic) on hypocotyl cell elongation. -Differentiate between the temporal and spatial effect 3. Study the role of auxins in the rearrangement of cortical microtubules during hypocotyl elongation -Use of GFP-expressor mutants and confocal microscopy to visualize the cell cytoskeleton -Extra: Study if the effect is consistent at different light intensities
