Summary: Dynamics of xylem formation and development in six juvenile and six mature trees of Douglas fir (Pseudotsuga menziesii (Mirb.) Franco) planted in the Czech Republic will be monitored. It is required to take samples (microcores) repetitively in a week interval during the growing season by means of the Trephor tool. Microcores will be cut on a rotary microtome to make permanent microslides of transversal sections. Number of cells in differentiation zones will be counted for each day of sampling. Results will show differences in wood formation dynamics of trees of different age (juvenile and mature).
